Description: This antibody recognizes a cell surface glycoprotein of 80-95 kDa (CD44) on lymphocytes, monocytes, and granulocytes (Leucocyte Typing Workshop V). Its epitope is resistant to digestion by trypsin and chymotrypsin. The CD44 family of glycoproteins exists in a number of variant isoforms, the most common being the standard 85-95 kDa or hematopoietic variant (CD44s). Higher molecular weight isoforms are described in epithelial cells (CD44v), which are believed to function in intercellular adhesion and stromal binding. CD44 immunostaining is commonly used for the discrimination of urothelial transitional cell carcinoma in-situ from non-neoplastic changes in the urothelium.

CF® dyes are Biotium's next-generation fluorescent dyes. CF®405S is a blue fluorescent dye (Ex/Em 404/431 nm) with superior brightness compared to other blue dyes; it is also compatible with super-resolution imaging by SIM. Description: This antibody recognizes a transcription factor of 64-67 kDa, identified as c-myc. Its epitope spans aa 410-419 (EQKLISEEDL), which is a specific portion of an alpha helical region of human c-myc protein. This MAb shows no cross-reaction with v-myc. c-myc is involved in the control of cell proliferation and differentiation and is amplified and/or overexpressed in a variety of tumors. Overexpression of c-myc protein occurs frequently in luminal cells of prostate intraepithelial neoplasia as well as in most primary carcinomas and metastatic disease.

CF® dyes are Biotium's next-generation fluorescent dyes. CF®488A is a green fluorescent dye (Ex/Em 490/515 nm) with excellent brightness and photostability. The dye is minimally charged for less non-specific binding. CF®488A also is compatible with super-resolution imaging by TIRF.

Description: In Western blotting, this antibody detects an antigen of 125 kDa in human liver and 135 kDa in tumors of histiocytic origin. Comparative study of this MAb and a standard CD68 MAb showed that their antigens are different. Its antigen in all macrophage types studied is located on the plasma membrane and within cytoplasmic structures including lysosomes. This MAb shows a restricted reactivity to cells of the monocyte/macrophage system. It specifically reacts with blood monocytes and stains resident macrophages in a wide variety of human tissues. This MAb does not stain antigen-presenting cells, e.g., Langerhans cells. Reportedly, its reactivity is restricted to histiocytes and macrophages.

Description: This antibody recognizes a phosphor-protein of 45 kDa, identified as MyoD1. This MAb does not cross react with myogenin, Myf5, or Myf6. Antibody to MyoD1 labels the nuclei of myoblasts in developing muscle tissues. MyoD1 is not detected in normal adult tissue, but is highly expressed in the tumor cell nuclei of rhabdomyosarcomas. Occasionally nuclear expression of MyoD1 is seen in ectomesenchymoma and a subset of Wilm s tumors. Weak cytoplasmic staining is observed in several non-muscle tissues, including glandular epithelium and also in rhabdomyosarcomas, neuroblastomas, Ewing s sarcomas and alveolar soft part sarcomas.

Description: This antibody recognizes an 85-115 kDa protein (variation with cell type), identified as intercellular adhesion molecule (ICAM-1) (Workshop IV). It has 7 potential N-linked glycosylation sites. ICAM-1 is a single chain glycoprotein of Ig supergene family, present on unstimulated endothelial cells (EC) and on a variety of other cell types including activated fibroblasts, EC, macrophages, and lymphocytes. ICAM-1 mediates cell adhesion by binding to integrins CD11a/CD18 (leukocyte adhesion molecule, LFA-1) and to CD11b/CD18 (Mac-1). This interaction enhances antigen-specific T-cell activation. ICAM-1 also binds to CD43 and to Plasmodium falciparum infected RBCs. W-CAM-1 MAb blocks aggregation of cell lines mediated by the ICAM-1 and blocks homotypic binding of purified populations of activated T- and B-lymphocytes and also aggregation of mixed T- and B-cell blasts. It inhibits T-cell adhesion to normal human endothelial cells. Activation induced by cell-cell contact (mixed lymphocyte reaction, T-cell mediated B-cell activation) is significantly inhibited. This MAb blocks elements of both effector arms of immune system (cytotoxic cell function and Ig production).

Description: This antibody recognizes an N-glycosylated glycoprotein of 120 kDa with intra-chain disulfide bonds, identified as CD50 or ICAM-3. CD50 is the major ligand for LFA-1 (CD11a/CD18) and may have signaling role to increase adhesion.
