Description: HLA-B belongs to the HLA class I heavy chain paralogues. This class I molecule is a heterodimer consisting of a heavy chain and a light chain (beta-2 microglobulin). The heavy chain is anchored in the membrane. Class I molecules play a central role in the immune system by presenting peptides derived from the endoplasmic reticulum lumen. They are expressed in nearly all cells. The heavy chain is approximately 45 kDa and its gene contains 8 exons. Exon 1 encodes the leader peptide, exon 2 and 3 encode the alpha1 and alpha2 domains, which both bind the peptide, exon 4 encodes the alpha3 domain, exon 5 encodes the transmembrane region and exons 6 and 7 encode the cytoplasmic tail. Polymorphisms within exon 2 and exon 3 are responsible for the peptide binding specificity of each class one molecule. Typing for these polymorphisms is routinely done for bone marrow and kidney transplantation. Hundreds of HLA-B alleles have been described.

Description: This MAb reacts with a 200 kDa protein, identified as heavy sub-unit of neurofilaments (NF-H). Neurofilaments make up the main structural elements of axons and dendrites and are found in neurons, peripheral nerves, and sympathetic ganglion cells. Neurofilaments consist of three major subunits with molecular weights of 68 kDa (NF-L), 160 kDa (NF-M) and 200 kDa (NF-H). Anti-neurofilament stains a number of neural, neuroendocrine, and endocrine tumors. Neuromas, ganglioneuromas, gangliogliomas, ganglioneuroblastomas, and neuroblastomas stain positively for anti-neurofilament. Neurofilaments are also present in paragangliomas as well as adrenal and extra-adrenal pheochromocytomas. Carcinoids, neuroendocrine carcinomas of the skin, and oat cell carcinomas of the lung also express neurofilament.

CF® dyes are Biotium's next-generation fluorescent dyes. CF®647 is a far-red fluorescent dye (Ex/Em 650/665 nm) with excellent brightness. It also is compatible with super-resolution imaging by STORM.

Description: This antibody reacts with a 75 kDa melanocyte-specific gene product, identified as Tyrosinase-related protein-1 (TRP-1). It is involved in melanin synthesis. TRP1 is present on the melanosomal membranes of melanoma, normal melanocytes and nevi.Recent evidence suggests that TRP-1 is involved in maintaining stability of tyrosinase protein and modulating its catalytic activity. TRP-1 is also involved in maintenance of melanosome ultrastructure and affects melanocyte proliferation and cell death.

Description: This antibody recognizes a protein of 185 kDa, which is identified as c-erbB-2/HER-2/neu. Its epitope is localized in the extracellular domain. C-erbB-2/HER-2 is a member of the EGFR family. This MAb is specific and shows minimal cross-reaction with other members of the EGFR-family. Receptors of this family are located on the plasma membrane and consist of an extracellular ligand-binding domain that is connected to a large intracellular domain by a single transmembrane sequence. c-erbB-2/HER-2 protein is over-expressed in a variety of carcinomas especially those of breast and ovary.

CF® dyes are Biotium's next-generation fluorescent dyes. CF®594 is a deep red fluorescent dye (Ex/Em 593/614 nm). It yields the brightest conjugates among spectrally similar dyes, and has excellent photostability.

Description: This MAb recognizes the HLA-B27 cell surface antigen on human cells. It may be used to HLA type human lymphocytes. Approximately 60% of patients with ankylosing spondylitis are HLA-B27 positive. This reagent can be used to help identify this HLA haplotype in human lymphocytes. Major histocompatibility complex (MHC) molecules form an integral part of the immune response system. They are cell-surface receptors that bind pep- tides and present them to T lymphocytes. Human leukocyte antigens (HLAs) are polymorphic members of the MHC family that are specifically involved in the presentation of antigens to the T cell receptor. There are two classes of HLA antigens: class I (HLA-A, HLA-B and HLA-C) and class II (HLA-D). Class I molecules are expressed in nearly all cells and play a central role in the immune system by presenting peptides derived from the endoplasmic reticulum. The differential structural properties of MHC class I and class II molecules account for their respective roles in activating different populations of T lymphocytes. HLA-B encodes a membrane anchored heavy chain, which hetero-dimerizes with a light chain (Beta-2-Microglobulin) to form MHC-I. Polymorphisms yield hundreds of HLA-B alleles. The HLA-B27 allele appears with increased frequency in uveitis patients.

Description: This antibody recognizes an epitope on the extracellular domain of gonadotropin releasing hormone (GnRH) receptor or luteinizing hormone receptor (LHCGR). Lutropin (also designated luteinizing hormone) plays a role in spermatogenesis and ovulation by stimulating the testes and ovaries to produce steroids. Gonadotropin (also designated choriogonadotropin) production in the placenta maintains estrogen and progesterone levels during the first trimester of pregnancy. Ovaries and testes abundantly express luteinizing hormone/choriogonadotropin receptor. GnRH receptor contains seven hydrophobic transmembrane domains connected by hydrophilic extracellular and intracellular loops characteristic of G-protein coupled receptors. GnRH stimulates the gonadotrophs of the anterior pituitary to secrete luteinizing hormone (LH) as well as follicle-stimulating hormone (FSH). GnRH influences the protective effect of pregnancy and Gonadotropin against breast cancer. The expression of GnRH on breast carcinoma correlates in part to the degree of tumor differentiation. GnRH-positive breast tumors occur more frequently in tumors with greater cell differentiation in premenopausal women. GnRH is present in luteal and granulosa cells as well as in ovarian cell membrane preparations.

Description: This MAb reacts with a protein of 22 kDa, identified as β sub-unit of HCG. It does not cross react with the α sub-unit. HCG is a glycoprotein, which is secreted in large quantities by normal trophoblasts. It is present only in trace amounts in non-pregnant urine and sera but rises sharply during pregnancy. HCG is composed of two non-identical, non-covalently linked polypeptide chains designated as the alpha and beta subunits. The beta subunit is identical to that of thyroid stimulating hormone (TSH), follicle stimulating hormone (FSH), and luteinizing hormone (LH). hCG MAb detects cells and tumors of trophoblastic origin such as choriocarcinoma. Large cell carcinoma and adenocarcinoma of the lung demonstrate anti-hCG positivity in 90% and 60% of cases respectively. 20% of lung squamous cell carcinomas are positive. hCG expression by non-trophoblastic tumors may indicate aggressive behavior.
