Description: Acrylamide is a monomer used in a variety of synthetic processes to form polymers and copolymers. It polymerizes in the presence of free radicals in aqueous solutions. It is usually used to prepare polyacrylamide gels for electrophoresis separation of biomolecules. Common reaction initiators are riboflavin and ammonium persulfate; varying the ratio of acrylamide to crosslinking agent permits the formation of a gel with predictable average pore size and texture. Acrylamide tends to hydrolyze under acidic or basic conditions to form acrylic acid. Polyacrylamide plastics have numerous commercial applications, due in part to the tendency of polyacrylamide to adsorb many times its own mass in water. Acrylic acid and any ionic impurities in the acrylamide can have significant effects on the performance of the acrylamide gel formed, since the voltage across the gel is affected by the ionic charge of the gel and usage buffers.

Description: A vitamin/enzyme cofactor found in milk, eggs, malted barley, liver, kidney, heart, leafy vegetables. Richest natural source is yeast. Occurs in the free form only in the retina of the eye, in whey, and in urine; bioactive forms occurring in tissues and cells are riboflavin monophosphate and flavine-adenine dinucleotide. Used as a photoinitiator for polymerization of polyacrylamide gels. It forms free radicals in aqueous solution in the presence of light. Riboflavin photodecomposes to leucoflavin. No free radicals are formed in the absence of oxygen but traces of oxygen allows for leucoflavin to reoxidize with free-radical generation. The catalysts TEMED or DMAPN are commonly added to speed up the free radical formation. The free radicals will cause acrylamide and bis-acrylamide to polymerize to form a gel matrix which can be used for electrophoresis. Riboflavin is commonly used in the stacking gel for non-denaturing polyacrylamide electrophoresis because native proteins can be senstive to persulfate ions from ammonium persulfate. Another advantage of riboflavin over ammonium persulfate is that it will not start polymerization until the gel is illuminated. Riboflavin causes less denaturing than other initiators. Gel strength can be improved when used with PTO (Pyrithione). Soluble in 0.1 M Sodium Hydroxide (50 mg/ml - clear, yellowish-orange solution); only slightly soluble in water.
