Description: CENPW was initially identified as a gene that was upregulated in multiple cancers, and whose overexpression in mouse fibroblast cells gave rise to distinct cancer-specific phenotypes. It was later found to be a nuclear protein that associated with CENPT, a component of CENPA nucleosome complex in the centromere, and is required for proper kinetochore function. CENPW also specifically interacts with the nucleolar phosphoprotein nucleophosmin, also known as B23 It has been suggested that nucleophosmin functions in the assembly of the kinetochore by interacting with CENPW during interphase. Overexpression of CENPW in the SKOV-3 human ovarian cancer cell line as well as in the zebrafish embryo led to apoptosis, suggesting that high levels of CENPW induces apoptotic cell death.

Description: Cyclic AMP-dependent transcription factor ATF-4 (ATF4) is a basic leucine-zipper (bZip) transcription factor, which regulates amino acid metabolism, DNA damage repair, chromatin remodeling, and apoptosis in response to cellular and ER stress. ATF4 works with various proteins, such as C/EBP homology protein (CHOP), aspargine synthetase (ASNS), and cAMP response element (CRE) among others to mediate cellular stress. ATF4 also regulates glucose homeostasis by suppressing beta-cell proliferation and insulin production. Furthermore, ATF4 targets the histone demethylase JMJD3 to alter chromatin structure and enhance gene transcription in response to amino acid deprivation. Anti-ATF4 is ideal for researchers interested in Cell Signaling, Oncology, Cell Differentiation, and Apoptosis; relevant pathways include MAPK signaling pathways, Activation of cAMP-Dependent PKA, CREB pathways, GPCR pathways and Rho Family GTPases.

Description: DR5 antibody detects human DR5 precursor. Apoptosis is induced by certain cytokines including TNF and Fas ligand in the TNF family through their death domain containing receptors. TRAIL/Apo2L is a new member of the TNF family. DR4 was recently identified as the receptor for TRAIL. A novel death domain containing receptor for TRAIL was more recently identified and designated DR5, Apo2, TRAIL-R2, TRICK2, or KILLER by several groups independently. Like DR4, DR5 transcript is widely expressed in normal tissues and in many types of tumor cells. DR5 binds to TRAIL and mediates TRAIL induced cell death. Overexpression of DR5 induces apoptosis and activates NF-alpha;B. Anti-DR5 antibodies are ideal for investigators involved in NFkappaB, Apoptosis, Cytokines and Growth factor research.

Description: EFCAB4B, also known as Calcium release-activated calcium channel regulator 2A, is a novel Ca2+-binding EF-hand protein that is thought to play a key role in store-operated Ca2+ entry in T-cells by regulating CRAC channel activation. EFCAB4B acts as a cytoplasmic calcium-sensor that forms a complex with ORAI1 and STIM1 at the junctional regions between the plasma membrane and the endoplasmic reticulum upon low Ca2+ concentration. A closely related protein, EFCAB4A, is likely to play a similar role as EFCAB4B, but the detailed function of EFCAB4A is still under investigation.

Description: Dendritic cells (DCs) that control immune responses were recently found to capture and transport HIV from the mucosal area to remote lymph nodes, where DCs hand over HIV to CD4+ T lymphocytes. DCs also amplify the amount of virus and extend the duration of viral infectivity. Multiple strains of HIV-1, HIV-2 and SIV bind to DCs via DC-SIGN. ICAM-3 is the natural ligand for DC-SIGN. A DC-SIGN homologue (termed DC-SIGNR, L-SIGN, and DC-SIGN2) was identified recently. DC-SIGN forms a novel gene family with DC-SIGNR and many alternatively spliced isoforms of DC-SIGN and DC-SIGNR. The expression of DC-SIGN was found in mucosal tissues including placenta, small intestine, and rectum.

Description: Cyclin O, also known as CCNO, has recently been identified as a Cdk1- and Cdk2-activating cyclin specific to apoptosis in lymphoid cells. Cyclin O binds to and activates Cdk2 in response to instrinsic apoptotic stimuli such as glucocorticoids or DNA-damaging agents. Supression of Cyclin O expression by shRNA leads to the inhibition of glucocorticoid and DNA-damage-induced apoptosis due to a failure of apical caspase activation while leaving the CD95 death receptor-mediated apoptosis intact. Note: this gene, which had a previous symbol of UNG2, was erroneously identified as a uracil DNA glycosylase. A later publication identified this gene's product as a cyclin protein family member.

Description: Adhesion to extracellular matrix regulates cell survival through both integrin engagement and appropriate cell spreading. Anoikis is the molecular mechanism of apoptosis induced by integrin detachment. Bit1 (Bcl-2 inhibitor of transcription 1) was recently identified as being involved in this process. Bit1 is a mitochondrial protein that is released into the cytoplasm upon onset of apoptosis where it forms a complex with AES, a small Groucho/transducin-like enhancer of split (TLE) protein and induces caspase-independent apoptosis. Both AES and TLE proteins are transcriptional co-repressors that play important roles in neurogenesis, segmentation, and sex determination. It has been suggested that Bit1-AES complexes turn off a survival-promoting gene transcription program controlled by TLE. Interestingly, apoptosis of cells transfected with Bit1 and AES could be inhibited if the cells were allowed to attach to fibronectin through the alpha5beta1 integrin suggesting that the Bit1-AES pathway contributing to anoikis is regulated by integrins, and in particular, the alpha5beta1 integrin.

Description: The Betatrophin protein, also known as the hepatocellular carcinoma-associated protein TD26, is an adipocyte- and liver-enriched insulin targeted gene that is thought to play a role in lipid metabolism be regulating serum triglyceride levels (1,2). Betatrophin is closely related to the angiopoietin-like (ANGPTL) protein family. Betatrophin mRNA increases in the cold environment and after treatment with a high-fat diet, while its mRNA levels decrease in response to fasting, suggesting that it may play a role in mediating thermogenesis and energy homeostasis (3).

Description: Bnip3L antibody detects human origin. Members in the Bcl-2 family are critical regulators of apoptosis by either inhibiting or promoting cell death. Bcl-2 homology 3 (BH3) domain is a potent death domain. BH3 domain containing pro-apoptotic proteins, including Bad, Bax, Bid, Bik, Hrk, Nip3, and Bim, form a growing subclass of the Bcl-2 family. A novel BH3 domain containing protein was recently identified and designated Bnip3L, Bnip3alpha, and Nix (for Nip3-like protein X). Bnip3L/Bnip3alpha/Nix is a homolog of the E1B 19K/Bcl-2 binding and pro-apoptotic protein Bnip3. Overexpression of Bnip3L induces apoptosis. Bnip3L interacts with and overcomes suppresses by Bcl-2 and Bcl-xL. Bnip3L is localized in mitochondria. The messenger RNA of Bnip3L is ubiquitously expressed in human tissues. Bnip3L and Bnip3 form a new subfamily of the pro-apoptotic mitochondrial proteins. Anti-Bnip3L antibodies are ideal for investigators involved in Apoptosis research.

Description: Bcl9L, a homolog of Bcl9, was initially identified through a bioinformatics screening. It is expressed in fetal brain, adult lung, eye and prostate, in addition to several types of tumors including pancreatic and prostate cancers. Bcl9L has been shown to interact with beta-catenin, a target of the Wnt signaling pathway, and is required for enhanced beta-catenin-T-cell factor (TCF)-mediated transcription in colorectal tumor cells, possibly by translocating beta-catenin to the nucleus. Other studies have indicated that Bcl9L expression correlates with high nuclear grade cancer phenotype and the expression of ErbB2/HER-2 in breast cancers, suggesting that activity may occur in other types of cancer. Bcl9L has also been shown to be critical for Wnt-mediate regulation of stem cell traits in colon epithelium and adenocarcinomas which are associated with tumor invasion, metastasis, and resistance to therapy.

Description: Actins are highly conserved proteins that are involved in cell motility, structure and integrity, processes that are crucial for tissue development and the development of organism. The actin cytoskeleton is one of the principal drivers of cell motility and is capable of responding to complex signaling cascades. Recent evidence suggests that it may play key roles in regulating apoptosis and aging. Beta actin is one of six different actin isoforms which have been identified. Like GAPDH, beta-actin is constitutively expressed at high levels in almost all tissues and cell lines making it ideal for use as a loading control marker in immunoblots.

Description: DARC, also known as the Duffy antigen/chemokine receptor, is a seven-transmembrane protein homologous to the classical chemokine G-protein coupled receptors (GPCRs) with the exception of the motif required for G protein coupling. DARC can bind with high affinity several chemokines without transducing any signal, suggesting it may modulate the signals normally induced by these chemokines. Recently, DARC was found to interact with KAI1, a four transmembrane protein recently identified as a tumor metastasis suppressor protein. It is thought that tumor cells dislodged from the primary tumor and expressing KAI1 interact with DARC proteins expressed on vascular cells, transmitting a senescent signal to the tumor cells, while tumor cells that have lost KAI1 expression can proliferate and potentially give rise to metastases. At least three isoforms of DARC are known to exist.
