Description: OCC-1 (Overexpressed in colon carcinoma-1) was initially identified as a novel human cDNA overexpressed in a colon carcinoma cell line. Elevated levels of OCC-1 mRNA were observed in three of eight colon carcinomas, suggesting that OCC-1 may be a hallmark of only a subset of colon carcinomas. It is highly expressed in skeletal muscle, kidney, placenta, and pancreas tissues, with low to no expression in heart, brain, lung and liver tissues. At least two different OCC-1 mRNA species are known to exist.

Description: The ORMDL (ORM1-like) family of proteins consist of ORMDL1, ORMDL2 and ORMDL3 that may play roles in gamma-secretase complex maturation. Localized to the membrane of the endoplasmic reticulum, ORMDLs are widely expressed and are implicated in the expression of Presenilin (PSI) which is involved in Alzheimer's disease (AD). Expression of ORMDL1 is down-regulated in PSI mutations, suggesting a possible role as a therapeutic target for AD. ORMDLs have recently been identified as a potential risk factor for childhood asthma.

Description: The nuclear pore complex (NPC) is a protein assembly localized at the nuclear rim and mediates macromolecular transport between the nucleus and the cytoplasm. The mammalian nucleoporin (NUP)-160 is part of the hetero-oligomeric complex that also contains NUP107, NUP133, NUP96, and mammalian homolog of yeast sec13p. While the majority of the NUP107-160 nuclear pore sub-complex localizes to the nuclear pore, a small fraction is observed at kinetochores and pro-metaphase spindle poles in mitotic cells in association with proteins such as Mad1, Mad2, Bub3 and Cdc20. Immunodepletion of the NUP107-160 complex resulted in defective spindle assembly indicating that it has multiple functions. NUP160 has recently been identified as an HIV dependency factor (HDF), suggesting that NUP160 may be an important drug target in HIV treatment. Multiple isoforms of NUP160 are known to exist.

Description: MARVELD1, (MARVEL domain-containing 1), is one of the MARVEL (MAL and related proteins for vesicle trafficking and membrane link) domain-containing proteins. It is a nuclear-localized protein that is widely expressed in normal human tissues. While it has been reported to be overexpressed in colon cancer, MARVELD1 protein levels are generally lower in several primary tumors including ovary, breast, kidney, bladder and liver. This lower level of MARVELD1 expression in tumors is thought to be due to a higher methylation status of the MARVELD1 promoter as treatment of MDA-MB-361 with 5-aza-2'-deoxycytidine restores MARVELD1 expression, suggesting that loss of MARVELD1 expression may contribute to tumorgenicity.

Description: The 50 kDa type III intermediate filament protein glial fibrillary acidic protein (GFAP) is a major structural component of astrocytes. GFAP associates with the calcium binding protein annexin II-p2 and S-100. Association with these proteins together with phosphorylation regulates GFAP polymerization. Astroycytes respond to brain injury by proliferatin (astrogliosis), and one of the first events to occur during astrocyte proiliferation is increased GFAP expression. Interestingly, antibodies to GFAP have been detected in individuals with dementia. Anti-GFAP is ideal for investigators involved in Neuroscience Research, including Alexander Disease, Oligodendroglioma, Cytoskleton Remolding Neurofilaments and PIP3/AKT Signaling.

Description: Accumulation of the amyloid-beta (Abeta) plaque in the cerebral cortex is a critical event in the pathogenesis of Alzheimer's disease. Abeta peptide is generated by proteolytic cleavage of the beta-amyloid protein precursor (APP) at beta- and gamma-sites by two proteases. APP is first cleaved by beta-secretase, producing a soluble derivative of the protein and a membrane anchored 99-amino acid carboxy-terminal fragment (C99). The C99 fragment serves as substrate for gamma-secretase to generate the 4 kDa amyloid-beta peptide, which is deposited in the brains of all suffers of Alzheimer's disease. The long-sought beta-secretase was recently identified by several groups independently and designated beta-site APP cleaving enzyme (BACE) and aspartyl protease 2 (Asp2). BACE/Asp2 is a novel transmembrane aspartic protease and colocalizes with APP.

Description: Growth differentiation factors (GDFs) are members of the transforming growth factor (TGF) superfamily that is involved in embryonic development and adult tissue homeostasis. Both GDF6 and GDF7 are closely related to GDF5 which has been shown to induce activation of plasminogen activator, thereby inducing angiogenesis. It is predominantly expressed in long bones during fetal embryonic development and is involved in bone formation. In Xenopus, GDF6 is expressed at the edge of the neural plate and within the anterior neural plate including the eye fields. GDF6 is required for normal formation of some bones and joints in the limbs, skull, and axial skeleton. It may regulate patterning of the ectoderm by interacting with bone morphogenetic proteins (BMPs), and control eye development. Mutations in this gene result in colobomata, which are congenital abnormalities in ocular development, and in Klippel-Feil syndrome (KFS), a congenital disorder of spinal segmentation.

Description: BATF is a nuclear basic leucine zipper protein that belongs to the AP-1/ATF superfamily of transcription factors. The leucine zipper of this protein mediates dimerization with members of the Jun family of proteins. This protein is thought to be a negative regulator of AP-1/ATF transcriptional events and blocks cellular transformation by Ras and Fos. BATF also is required for the differentiation of IL-17-producing T helper (TH17) cells, a CD4+ subset of T cells that coordinates the inflammatory response in host defense. In both T cells and B cells, BATF is required for the appropriate regulation of the class-switch recombination.

Description: Apoptosis plays a major role in normal organism development, tissue homeostasis, and removal of damaged cells. Disruption of this process has been implicated in a variety of diseases such as cancer. The Bcl-2 family of proteins is comprised of critical regulators of apoptosis that can be divided into two classes: those that inhibit apoptosis and those that promote cell death. Bak, a pro-apoptotic Bcl-2 family member, is an oligomeric protein that localizes to the mitochondria. It is thought to share significant functional homology with Bax, another pro-apoptotic Bcl-2 family member, as disruption of bak or bax has little effect on cell death, but mice lacking both genes display multiple developmental defects and cells lacking bak and bax show decreased apoptotic capability.

Description: BAPX1 is the mammalian homolog of the Drosophila bagpipe homeobox gene and is expressed in the splanchnic mesoderm and embryonic skeleton. It is one of the earliest developmental markers for the sclerotome portion of the somite and the gut mesentery. BAPX1 is required for normal skeletal development; homozygous inactivating mutations in the BAPX1 gene result in spodylo-megaepiphyseal-metaphyseal dysplasia (SMMD). It has also been suggested to play a role in the proper development of the mammalian gut and is required for distal stomach development as part of a BARX1-dependent pathway.

Description: BCAS2 was identified through differential display analysis as an mRNA species that was overexpressed in MCF7 and BT-20 breast cancer cell lines. The chromosomal region containing this gene, 1p13.3021, is amplified in these cells lines. BCAS2 is a transcriptional cofactor that enhances estrogen receptor-mediated gene expression, and directly interacts with the tumor suppressor p53 to reduce p53 transcriptional activity by reducing p53 protein level in the absence of DNA damage. Deprivation of BCAS2 through RNA inhibition induces apoptosis in p53-wild type cells, but causes G2-M arrest in p53-null or -mutant cells; this effect was reversed with the expression of ectopic BCAS2. BCAS2 may thus be potentially useful as a therapeutic target in the treatment of cancer.

Description: Members in the Bcl-2 family are critical regulators of apoptosis by either inhibiting or promoting cell death. Bcl-2 homology 3 (BH3) domain is a potent death domain. BH3 domain containing pro-apoptotic proteins, including Bad, Bax, Bid, Bik, and Hrk, form a growing subclass of the Bcl-2 family. A novel BH3 domain containing protein was recently identified and designated Bim or BOD in human, mouse and rat. Bim/BOD interacts with diverse members in the pro-survival Bcl-2 sub-family including Bcl-2, Bcl-xL and Bcl-w. Bim/BOD induces apoptosis. The messenger RNA of Bim is ubiquitously expressed in multiple tissues and cell lines.

Description: FAM59B is related to the FAM59A protein (also known as GAREM). While little is known of the role of the FAM59B protein, FAM59A is a downstream molecule in the EGF signaling pathway and binds Grb2, an adaptor protein crucial to the transduction of growth signals from the plasma membrane to the nucleus. Like FAM59A, FAM59B may play a role in ligand-mediated signaling pathway of the EGF receptor and the tumorigenesis of cells.
