Description: TBC1D1 is the founding member of a family of proteins sharing a 180- to 200-amino acid TBC domain and presumed to have a role in regulating cell growth and differentiation. These proteins share significant homology with TRE2/USP6, yeast Bub2, and CDC16. TBC1D1 and TBC1D4 (AS160) have been demonstrated to be Rab GAPs (GTPase-activating proteins) that link upstream to Akt and phosphoinositide 3-kinase and downstream to Rabs involved in trafficking of GLUT4 vesicles. TBC1D1 regulates insulin-mediated GLUT4 translocation through its GAP activity, and is a likely Akt substrate. Mutations in the Tbc1d1 gene lead to some cases of severe human obesity.

Description: TCTN3 is a member of the Tectonic protein family, a group of evolutionarily conserved secreted and transmembrane proteins that regulate the Hedgehog (Hh)-mediated patterning of the neural tube. While the expression pattern and function of TCTN3 is not known, it is 58% similar to TCTN1. TCTN1 is expressed during embryonic development in regions that participate in Hh signaling, beginning in the gastrulation stages in the ventral node. Mice expressing mutant TCTN1 die between E13.5 and E16.5 and display holoprosencephaly, a defect associated with reduced Hh signaling, indicating the role of TCTN1 as an Hh activator. At later stages in development, TCTN1 is thought to also act as a repressor on the Hh pathway in the anterior and posterior neural tube.

Description: Growth hormone regulated TBC protein 1 (GRTP1) was identified through microarray analysis as a gene whose expression is regulated by growth hormone (GH). It contains the TBC signature motif of GTPase activator proteins of Rab-like small GTPases. GRTP1 mRNA is expressed at highest levels in testes, and is also present in kidney, lung, liver, and intestine. Administration of GH to mice resulted in an increase in GRTP1 mRNA in testes, but a decrease of GRTP1 mRNA in kidney and liver. GRTP1 was identified as an HIV dependency factor (HDF), suggesting that GRTP1 may be an important drug target in HIV treatment. Description: Tetratricopeptide repeat protein 5 (TTC5) is a member of a diverse group of functionally distinct proteins that are characterized by containing one or more tetratricopeptide repeats. Each motif consists of two anti-parallel a-helices such that tandem arrays of TPR motifs generate a right-handed helical structure with an amphipathic channel that may serve to accommodate the complementary region of a target protein. While the exact function of TTC5 remains unclear, it is thought that the TPR motifs serve to mediate protein-protein interactions such as those seen with protein chaperones HSP70 and HSP90 and some proteins involved in cell stress response signaling pathways such as protein phosphatase 5, suggesting that TTC5 may also function via protein-protein interactions mediated by its TPR motifs.
