Supplier: MP Biomedicals
Description: Acrylamide is a monomer used in a variety of synthetic processes to form polymers and copolymers. It polymerizes in the presence of free radicals in aqueous solutions. It is usually used to prepare polyacrylamide gels for electrophoresis separation of biomolecules. Common reaction initiators are riboflavin and ammonium persulfate; varying the ratio of acrylamide to crosslinking agent permits the formation of a gel with predictable average pore size and texture. Acrylamide tends to hydrolyze under acidic or basic conditions to form acrylic acid. Polyacrylamide plastics have numerous commercial applications, due in part to the tendency of polyacrylamide to adsorb many times its own mass in water. Acrylic acid and any ionic impurities in the acrylamide can have significant effects on the performance of the acrylamide gel formed, since the voltage across the gel is affected by the ionic charge of the gel and usage buffers.
Store at Room Temperature (15-30 °C), Dessicated.

Description: β-Mercaptoethanol can act as an enzyme reactivator in systems necessitating reduction for activation, and has been commonly used to reduce disulfide bonds in order to separate protein subunits for use in electrophoresis.
2-Mercaptoethanol is used to reduce disulfide linkages in solubilizing proteins for gel electrophoresis (typically used in SDS-PAGE sample buffer at 5% concentration). Also reduces excess oxidative polymerization of catalysts. Cleaving intermolecular (between subunits) disulfide bonds allows the subunits of a protein to separate independently on SDS-PAGE. Cleaving intramolecular (within subunit) disulfide bonds allows the subunits to become completely denatured so that each peptide migrates according to its chain length with no influence due to secondary structure. In solution, 2-mercaptoethanol is readily oxidized in air to a disulfide, especially at alkaline pH. Because of this property, it is widely used to protect proteins, enzymes in particular, from becoming inactive. An excess of 2-mercaptoethanol (generally used at 0.01 M) will maintain the protein thiol groups in their reduced state.
Supplier: Promega Corporation
Description: Available with green dye for direct-to-gel convenience, or in colorless format for applications where absorbance or fluorescence measurements are necessary prior to PCR cleanup
Catalog Number: (103010-566)
Supplier: Anaspec Inc
Description: The SensoLyte® Green Elastase Assay Kit is a FRET-based assay that detects elastase activity. The kit provides an elastin, natural substrate for elastases, labeled with 5-FAM fluorophore and QXL®520 quencher. Elastase-catalyzed hydrolysis yields brightly green fluorescence. Increase in fluorescence intensity is directly proportional to enzyme activity. The kit does not require any separation steps and can be used to continuously measure the kinetics of elastase.

Supplier: Promega Corporation
Description: The Reverse Transcription System provides reagents to efficiently reverse transcribe RNA into cDNA in fifteen minutes.
Catalog Number: (103010-144)
Supplier: Anaspec Inc
Description: The SensoLyte® Green Protease Assay Kit is widely used for detection of generic protease activities. The kit uses a casein derivative that is heavily labeled with a rhodamine derivative, resulting in almost total quenching of the conjugate's fluorescence. Protease-catalyzed hydrolysis relieves this quenching conjugate, yielding brightly green fluorescent dye-labeled peptides. The increase in fluorescence intensity is directly proportional to protease activity (Ex/Em=488/520 nm). The SensoLyte® protease assay kits do not require any separation steps and can be used to continuously measure the kinetics of a variety of exopeptidases and endopeptidases.

Supplier: Enzo Life Sciences
Description: The 90kDa molecular chaperone family includes 90kDa heat shock protein Hsp90 and 94kDa glucose-regulated protein grp94, both major molecular chaperones of the cytosol and the endoplasmic reticulum. Mammalian cells contain isoforms Hsp90α and Hsp90β, encoded by separate genes. The amino acid sequences of human and yeast Hsp90α are 85% and 90% homologous to those of Hsp90β, respectively. All known members of the Hsp90 protein family are highly conserved, especially in the N-terminal and C-terminal regions containing independent chaperone sites with different substrate specificity. These ubiquitous and highly conserved proteins account for 1-2% of all cellular proteins in most cells. Hsp90 functions as part of the cell’s powerful network of chaperones to fight the deleterious consequences of protein unfolding caused by non-physiological conditions. In the absence of stress, however, Hsp90 provides a necessary component of such fundamental cellular processes as hormone signalling and cell cycle control. In this context, researchers identified key regulatory proteins as substrates of Hsp90, including steroid receptors, cell cycle kinases involved in signal transduction, and p53. Hsp90 may act as a capacitor for morphological evolution by buffering widespread variation, potentially affecting morphogenic pathways. When temperature and other stress factors compromise Drosophilia Hsp90 buffering, cryptic variant expression occurs, and selection can lead to the continued expression of these traits, even after Hsp90 function is restored.
