Description: Fluor de Lys® Substrate is a highly sensitive and convenient alternative to radiolabeled, acetylated histones or peptide/HPLC methods for the assay of histone deacetylases. The Fluor de Lys® Substrate, which comprises an acetylated lysine side chain, is incubated with a sample containing HDAC activity (nuclear or cellular extract, purified enzyme, bead-bound immunocomplex, etc.). Deacetylation of the substrate sensitizes the substrate so that, in a second step, treatment with the Fluor de Lys® Developer produces a fluorophore. Histone deacetylases from all known classes have been successfully assayed with the Fluor de Lys® Substrate. These include human HDACs 1, 2, 3 and 8 (class I), human HDACs 4 and 9 (class II) and the sirtuins yeast Sir2, human SIRT1 and human SIRT2 (class III; reaction must include the sirtuin substrate NAD+). The substrate is cell-permeable and serves as the basis for the HDAC Cellular Activity Assay Kit (Prod. No. BML-AK503). NOTE: Must be used in conjunction with Fluor de Lys® Developer (Prod. No. BML-KI105). Sufficient for 100-1000 assays of HDACs or sirtuins (50 µl reaction volume, 50-500 µM (substrate).

Description: BIOMOL Green™ provides a simple and convenient method for colorimetric phosphate quantitation (abs. 600-680 nm). Unlike other molybdate/malachite green-based assays, BIOMOL Green™ doesn’t require multiple solutions or reagents prepared fresh on the day of the assay. BIOMOL Green™ is extremely stable (>6 mos. at 4°C) and is simply mixed, at room temperature, with any enzymatic reaction that has released free phosphate. Reported applications include assays for phospholipid phosphatases, tyrosyl-tRNA synthetase (coupled with pyrophosphatase) and a viral RNA triphosphatase.BIOMOL Green™ has perhaps been most widely applied to protein phosphatase assays. Typically, a protein phosphatase (e.g. PTP1B or LAR) is simply incubated with an appropriate phosphopeptide (e.g. IR5,9,10). Addition of BIOMOL Green™ stops the reaction and begins color development, which is read 20-30 min. later.BIOMOL Green™ may be used in cuvette or microplate-based assays and is ideal for high-throughput applications. Each kit comes with a standardized phosphate solution for assay calibration.

Description: RNA-Solv® Reagent is a one reagent system for the isolation of total RNA from cells and tissues. The reagent, a single-phase solution consisting of phenol and guanidine isothiocyanate, is a modification of the single-step RNA isolation method developed by Chomczynski and Sacchi. The sample is homogenised and lysed in RNA-Solv® Reagent, which maintains the integrity of the RNA while disrupting and denaturing endogenous RNases and other cellular components. Extraction of the lysate with chloroform further denatures proteins and separates the mixture into an organic and an aqueous phase. RNA remains exclusively in the aqueous phase, and is subsequently recovered by isopropanol.
