VSV Pseudotyped RSV Fusion with Luciferase Reporter (A2 strain)

This pseudotyped virus uses recombinant vesicular stomatitis virus (rVSV) to carry the full length fusion (F) protein of Human respiratory syncytial virus (RSV) isolate A2 strain (UniProtKB/Swiss-Prot: P03420.1, with prefusion-stabilizing mutations). RSV is a respiratory virus with fusion (F) protein as the major target of neutralizing antibodies/vaccine-elicited sera.

As pseudovirus infectivity of rVSV without its original G is restricted to a single round of replication, the pseudotypes can be handled using BSL-2 containment practices. Infection of cells with this pseudotyped virus carrying luciferase reporter results in high level luciferase activity. Titration result showing that the starting 2-fold diluted pseudovirus generates signal ~10000-fold higher than uninfected control (cell alone as background in blue). Three known neutralizing mAbs (D25, 101F, and MPE8) were used to validate its neutralizing activity against this pseudovirus.
A complete protocol is supplied with the shipping of product. Briefly, incubate 50 μl diluted virus with sample in each well of 96-well plate for 60 min at 37 °C. Then add 50 μl target cells, such as HEp-2, Vero, or others expressing RSV receptor, followed by luciferase signal reading.
Note: Pseudoviruses are intended for research use only. A positive control mAb is included with pseudovirus purchase.
Envase: Shipped with dry ice. Requires −80 °C storage. Multiple freeze/thaw cycles will reduce sensitivity. Recommend only 1 cycle. Aliquot after the first thaw.
Precaución: Handle in biosafety cabinet in BSL-2. Contacted tips and tubes should be decontaminated by 10% disinfecting bleach. For research use only.
