This pseudotyped virus uses recombinant vesicular stomatitis virus (rVSV) to carry the full length fusion (F) protein of Human metapneumovirus (HMPV) isolate NL/1/99 B1 strain (GenBank: AY525843.1). HMPV is a respiratory virus with fusion (F) protein as the sole target of neutralizing antibodies. As pseudovirus infectivity of rVSV without its original G is restricted to a single round of replication, the pseudotypes can be handled using BSL-2 containment practices.

Description: Infection of cells with this pseudotyped virus carrying luciferase reporter results in high level luciferase activity. See our titration result showing that the starting 2-fold diluted pseudovirus generates signal ~ 10,000-fold higher than uninfected control (cell alone as background in blue). An RSV and hMPV cross-reactive mAb RV35 (Cat # M72M26), along with 3 known neutralizing mAbs (101F, D25 and MPE8) were used to validate its neutralizing activity against this pseudovirus.


Biosafety level: Handle it in biosafety cabinet in BSL-2. Contacted tips and tubes should be decontaminated by 10% disinfecting bleach.


Use protocol: A complete protocol is supplied with the shipping of product. Briefly, incubate 50 μl diluted virus with your sample in each well of 96-well plate for 30 to 60 min at 37 °C. Then add 50 μl target cells, such as Vero E6 (J Clin Microbiol. 45: 1912-1919.), Lung cells or others expressing HMPV receptor, followed by luciferase signal reading.

Envase: Shipping with dry ice. Require −80 °C storage. Multiple freeze/Thaw cycles will reduce its sensitivity. Recommend only 1 cycle. Aliquot after the first thaw.
