Description: Camelids produce functional IgG isotypes that do not incorporate light chains. Comparative studies of old world camelids (Camelus bactrianus and Camelus dromedarius) and new world camelids (Lama pacos, Lama glama and Lama vicugna) have shown that heavy-chain-only immunoglobulins represent between 35% - 70% of total IgG in the sera of all species. At present, three subclasses of camelid IgG have been identified (IgG1, IgG2, IgG3), of which IgG2 and IgG3 lack the light chains. IgG1 binds strongly to protein A and G, is composed of conventional antibodies, and totals 25 % of serum IgG. Llama IgG1 antibodies migrate as a 150-kDa protein in SDS-polyacrylamide gel under non-reducing conditions. Under reducing conditions, IgG1 antibodies split into four proteins (two of each H- and L-chains), which gives protein bands of about 52-55 kDa (H-chain) and about 27 kDa (L-chain). Anti-Llama IgG1 generated in rabbit detects specifically the Llama IgG1 isotype. Description: p63 is a homolog of the tumor suppressor p53, it is identified in basal cells in the epithelial layers of a variety of tissues, including epidermis, cervix, urothelium, breast and prostate. p63, vital for the development of the prostate and selectively expressed by normal prostate basal cells, has been shown to be a promising complementary basal cell specific marker to High Molecular Weight-Cytokeratin (HMW-CK), for the differential diagnosis of benign prostatic lesions and prostatic carcinoma. p63 has also been shown to be a sensitive marker for lung squamous cell carcinomas (SqCC), with a sensitivity of ~90% . Description: p40 is a relatively unknown antibody that recognizes ΔNp63-a p63 isoform suggested to be highly specific for squamous/basal cells. Description: The HMB45 clone reacts with a neuraminidase-sensitive oligosaccharide side chain of a glycoconjugate present in immature melanosomes. The HMB45-reactive antigen is present in cutaneous melanocytes, prenatal and infantile retinal pigment epithelium and melanoma cells and is thought to be oncofetal in nature. This antibody has been shown to label the majority of melanomas. Clone A103 recognizes a protein of 20 kDa, identified as MART-1 (melanoma antigen recognized by T cells-1) or Melan-A. Melan-A is a useful addition to melanoma panels as it is apparently specific for melanocytic lesions. Studies have also shown that MART-1 is more sensitive than HMB45 when labeling metastatic melanomas. Tyrosinase is a key enzyme involved in the initial stages of melanin biosynthesis.
