Description: Accumulation of the amyloid-beta (Abeta) plaque in the cerebral cortex is a critical event in the pathogenesis of Alzheimer's disease. Abeta peptide is generated by proteolytic cleavage of the beta-amyloid protein precursor (APP) at beta- and gamma-sites by two proteases. APP is first cleaved by beta-secretase, producing a soluble derivative of the protein and a membrane anchored 99-amino acid carboxy-terminal fragment (C99). The C99 fragment serves as substrate for gamma-secretase to generate the 4 kDa amyloid-beta peptide, which is deposited in the brains of all suffers of Alzheimer's disease. The long-sought beta-secretase was recently identified by several groups independently and designated beta-site APP cleaving enzyme (BACE) and aspartyl protease 2 (Asp2). BACE/Asp2 is a novel transmembrane aspartic protease and colocalizes with APP.

Description: Growth differentiation factors (GDFs) are members of the transforming growth factor (TGF) superfamily that is involved in embryonic development and adult tissue homeostasis. Both GDF6 and GDF7 are closely related to GDF5 which has been shown to induce activation of plasminogen activator, thereby inducing angiogenesis. It is predominantly expressed in long bones during fetal embryonic development and is involved in bone formation. In Xenopus, GDF6 is expressed at the edge of the neural plate and within the anterior neural plate including the eye fields. GDF6 is required for normal formation of some bones and joints in the limbs, skull, and axial skeleton. It may regulate patterning of the ectoderm by interacting with bone morphogenetic proteins (BMPs), and control eye development. Description: DCLK3 is one of three doublecortin-like kinases similar to the Ca2+/calmodulin-dependent protein kinase (CaMK) family. DCLK3 mRNA, like that of the homologous DCLK1 and DCLK3, is highly expressed in adult brain, but only DCLK3 transcripts are present in liver and kidney, suggesting that DCLK3 may play other roles than in cortical development. Description: Damage-regulated autophagy modulator (DRAM) is a p53 target gene encoding a lysosomal protein that induces autophagy, a process that degrades cytosolic proteins and organelles. It has been suggested that activation of DRAM by p53 is simultaneous to the activation by p53 of one or more proapoptotic genes such as PUMA, Bax, etc., and that the signaling pathways regulated by these genes promote a full cell death response. Description: The conserved dopamine neurotrophic factor (CDNF) is a neurotrophic factor for dopaminergic neurons and highly homologous to the mesencephalic-astrocyte-derived neuro-trophic factor. Somewhat surprisingly, CDCF is expressed at higher levels in tissues such as heart, skeletal muscle and testes than in brain. Similar to the glial cell line-derived neurotrophic factor (GDNF), CDNF can prevent the 6-hydroxylamine (6-OHDA)-induced degeneration of dopaminergic neurons in a rat model of Parkinson's disease. Description: The cyclin-dependent kinase inhibitor 1C (CDKN1C), also known as p57KIP2, is a tight-binding, strong inhibitor of several G1 cyclin/CDK complexes and a negative regulator of cell proliferation. Mutations in this gene are implicated in sporadic cancers and Beckwith-Wiedemann syndrome, a cancer-predisposing syndrome, suggesting that this gene is a tumor suppressor candidate. CDKN1C is widely expressed during organogenesis with its expression declining after maturity. It is part of the CIP/KIP family of cyclin-dependent inhibitors (CKIs) which also includes CDKN1A and CDKN1B.
