Description: BX795 was initially developed as a PDK1 inhibitor (direct competitive inhibitor IC(50) 11nM for PDK1) and has been shown to be a potent and relatively specific inhibitor of TBK1 and IKK-epsilon. It blocks the phosphorylation, nuclear translocation, and transcriptional activity of interferon regulatory factor 3. BX795 also is a potent inhibitor of cell growth of multiple cancer cell lines with IC(50) values ranging from submicromolar amounts (0.368µM to greater than 450µM.

Description: Interleukin-23 receptor (IL-23R), is a member of the type I cytokine receptor family, type 2 subfamily. It is expressed by monocytes, Th1, Th0, NK and dendritic cells. Isoform 1 is specifically expressed in NK cells. IL-23 functions in innate and adaptive immunity and may participate in acute response to infection in peripheral tissues. IL-23 may be responsible for autoimmune inflammatory diseases and be important for tumorigenesis. IL-23R associates with IL-12RB1 to form the interleukin-23 receptor. It binds IL-23 and mediates T cells, NK cells and possibly certain macrophage/myeloid cells stimulation probably through activation of the Jak-Stat signaling cascade. Genetic variations in IL-23R are associated with inflammatory bowel disease type 17 (IBD17). IBD17 is a chronic, relapsing inflammation of the gastrointestinal tract with a complex etiology. Genetic variations in IL-23R also can cause susceptibility to psoriasis type 7.

Description: Human CD154 (CD40 ligand) is a member of the TNF family and is expressed on the surface of activated T cells. It can undergo proteolytic cleavage into a 19kDa immunologically active soluble form. Interaction of CD154 and CD40 is essential for isotype switching in B cells. Known genetic defects that alter this interaction lead to impaired immune system function. Increased levels of CD154 has been associated with autoimmune disorders including SLE, CLL and eosinophilic fasciitis. CD154 has been reported to be expressed on vascular endothelial cells, smooth muscle cells, macrophages and activated platelets indicating a role for the CD40-CD154 immunoregulatory signaling in artherosclerosis and cardiovascular disorders.

Description: Human CD152 (CTLA-4) is a cell surface glycoprotein expressed at low levels on activated T cells. CD152 is a high affinity receptor for the costimulatory molecules CD80 (B7-1) and CD86 (B7-2) and appears to function as a negative regulator of T cell activation. This product is a soluble fusion protein combining the extracellular (125aa) domain of human CD152 and murine IgG2a Fc (CTLA-4 Ig).

Description: Human CD95 (APO-1; Fas) is a type I cell surface glycoprotein that is strongly upregulated on activated T cells, B cells, NK cells and thymocytes. CD95 plays an important role in programmed cell death or apoptosis. Apoptosis appears to be a mechanism for regulating the immune response.

Description: Human CD166 is a type 1 glycoprotein expressed on activated T cells, B cells and monocytes and appears to be the ligand for CD6. Human CD166 may be important for activation of T cells.

Description: Human CD252 (CD134L; OX40 ligand) is a type II membrane protein with homology to TNF which is expressed on activated B cells and activated endothelial cells. CD252 binds to CD134 (OX40) present on activated T cells, providing a costimulatory signal. Blockade of this interaction in mouse abrogated immunological effects in several models of inflammation and rejection.

Description: RANKL is an osteoclast differentiation and activation factor. Augments the ability of dendritic cells to stimulate naive T-cell proliferation. Important regulator of interactions between T-cells and dendritic cells and may play a role in the regulation of the T-cell-dependent immune response. Plays an important role in enhanced bone-resorption in humoral hypercalcemia of malignancy.

Description: ACRP30 was identified as a novel adipocyte-specific synthesized and secreted protein with structural resemblance to complement factor C1q. Like adipsin, ACRP30 secretion is induced ~10-fold during adipocyte differentiation. Plasma levels are reduced in obese humans, and low levels are associated with insulin-resistance. Treatment of db/db mice with TZD increased ACRP30 levels.

Description: beta-PIX acts as a RAC1 guanine nucleotide exchange factor (GEF) and can induce membrane ruffling. It interacts with PAK kinases through the SH3 domain. It may function as a positive regulator of apoptosis and is involved in cell migration. It is expressed with high levels in the central nervous system.

Description: BAFF is a master regulator of peripheral B cell survival, and together with IL-6, promotes Ig class-switching and plasma cell differentiation. BAFF co-stimulates activated T cells. Increased levels of soluble BAFF have been detected in the serum of patients with various autoimmune diseases, such as Sjögren's syndrome, rheumatoid arthritis, multiple sclerosis and systemic lupus erythematosus (SLE). Furthermore, BAFF is found in inflammatory sites in which there is lymphoid neogenesis. BAFF levels are elevated in patients with multiple myeloma and B cell chronic lymphoid leukemia (B-CCL).

Description: AIM2 is a strong candidate as a tumor suppressor as it is a member of the HIN-200 (hemopoietic interferon-inducible, nuclear proteins containing a 200 amino acid repeat) family of proteins that have been shown to regulate cell growth and survival. The HIN-200 family of proteins consists of four members in human, IFI 16, MNDA, AIM2 and IFIXl. Recently it has been reported that AIM2 is a receptor for cytosolic dsDNA, which forms a novel inflammasome complex with ASC to activate caspase-1-mediated processing of IL-1beta.
