Description: FKBP3 contains 1 PPIase FKBP-type domain, belongs to the FKBP-type PPIase family. FK506- and rapamycin-binding proteins (FKBPs) constitute a family of receptors for the two immunosuppressants which inhibit T-cell proliferation by arresting two distinct cytoplasmic signal transmission pathways. FKBP3 is a cis-trans prolyl isomerase enzyme that binds the immunosuppressants FK506 and rapamycin, as well as histone deacetylases, the transcription factor YY1, casein kinase II, and nucleolin. It has a higher affinity for rapamycin than for FK506 and thus may be an important target molecule for immunosuppression by rapamycin.

Supplier: Bioss
Description: DDAH, a dimethylarginine dimethylaminohydrolase, hydrolyzes dimethyl arginine (ADMA) and monomethyl arginine (MMA), both inhibitors of nitric oxide synthases, and may be involved in in-vivo modulation of nitric oxide production (1,2). Impairment of DDAH causes ADMA accumulation and a reduction in cGMP generation (3). DDAH II, the predominant DDAH isoform in endothelial cells, facilitates the induction of nitric oxide synthesis by all-trans-Retinoic acid (atRA) (4). DDAH proteins are highly expressed in colon, kidney, stomach and liver tissues (1).

Description: Syntaxins are type II integral membrane proteins localized to membranes that regulate vesicle transport, protein maturation, and exocytosis. Syntaxin 1A resides in the nerve terminals of sensory neurons and nerve fibers reaching small blood vessels. Syntaxin 2 localizes to the apical plasma membrane and regulates epithelial-mesenchymal interactions. Syntaxin 6 is primarily localized to the trans-Golgi network, and partially co-localizes with AP-1 on clathrin-coated membranes. Syntaxin 13 is ubiquitously expressed and localizes to the tubular early and recycling endosomes, as well as endosomal vacuoles.

Description: FKBP25 is a member of the immunophilin protein family. The immunophilins are a highly conserved family of cis-trans peptidyl-prolyl isomerases that bind to and mediate the effects of immunosuppressive drugs, such as cyclosporin, FK506 and rapamycin. They inhibit T-cell proliferation by interrupting two specific cytoplasmic signal transmission pathways. FKBP25 is localized in the nucleus and is expressed in the brain, testis, ovary, and spleen. It may influence immunoregulation and cerebellum development, as well as protein folding and trafficking in neurons. FKBP25 associates with transcriptional repressor protein YY1 and histone deaceltylases, HDAC1 and HDAC2. FKBP25 may contain several casein kinase II phosphorylation sites, which are believed to be important for cell growth regulation.

Description: Lysosomal enzymes containing one or two mannose 6-phosphate (man6P) moieties are moved about in the cell by two distinct but interconnected cycles by means of 300 kDa cation-independent mannose 6-phospate receptors (MPR). MPR cycles and transports newly synthesized lysosomal enzymes between the TGN and late endosomes / early lysosomes. It also cycles and transports extracellular lysosomal enzymes between the plasma membrane and early endosomes via clathrin coated vesicles. The bulk of the MPR protein is in the extracellular/lumenal domain. The entire pool of MPRs cycles between these cellular compartments every 3 hours. The steady state distribution of MPR's is predominantly within late endosomes, fewer in the trans Golgi network and ~5-10 % at the cell surface. In addition to its man6P binding activity, the MPR contains a separate binding site for the type II insulin-like growth factor and is capable of binding both man6P and IGF-II simultaneously. An ~240 kDa soluble, truncated form, representing the extracellular domain of the protein, has also been found circulating in serum and is capable of binding both ligands.

Description: Common component of cell membranes, often as phosphatidyl conjugates. Is a potent (IC50=1-3 µM) and selective inhibitor of protein kinase C. Inhibition is competitive with diacylglycerol, phorbol dibutyrate and Ca2+ and it also inhibits PKC activation by other lipids. It acts on an equimolar basis with 1,2-dioleoylglycerol and does not affect other kinases such as MLCK and PKA. It is active in intact cells and has been identified as an endogenous constituent in HL-60 cells, neutrophils, rat liver, and brain and mouse tissues. Other activities include inhibition of phosphatidate phosphohydrolase, Na+,K+-ATPase, CTP:phosphocholine cytidylyltransferase, calmodulin-dependent enzymes, binding of factor VII to tissue factor, binding of thyrotropin releasing hormone to its receptor and activation of EGF receptor kinase, phospholipase D and casein kinase II.
