Description: This GLP-1 (7-36) amide peptide is fluorescently labeled at Tryptophan residue 25 with Fluorescein. This is the same type of fluorescent labeling as in Extendin-4 Flex peptide (Cat# AS-63899). GLP-1 (7-36) amide is an incretin hormone that causes glucose dependent release of insulin by pancreatic beta cells. It is the cleavage product of GLP-1 (1-36) amide peptide (cat# AS-22460). Both GLP-1 (7-36) and GLP-1 (7-37) - Cat# AS-20761, also play roles in gastric motility (gastric emptying), on the suppression of plasma glucagon levels (glucose production) and possibly on the promotion of satiety and stimulation of glucose disposal in peripheral tissues independent of the actions of insulin. GLP-1 (7-36) has a short half life of less than 2 minutes, and like GIP, is rapidly degraded by the enzyme dipeptidyl peptidase IV (DPP-4), which is widely expressed in a number of sites, including the endothelial cells of small gut arterioles. DPP-4 degrades GLP-1 (7-36) into the non insulinotropic GLP-1 (9-36) - Cat# AS-65070 (some studies suggest it may have weak insulinotropic activity). As a result, the majority of GLP-1 (and GIP) is inactivated as an insulinotrope before reaching the systemic circulation.

Description: Recognizes the blood group H type 2 antigens, trisaccharide Fuc(a1-2)Gal(b1-4)GlcNAc(b1) of human origin. This protein is the basis of the ABO blood group system. The histo-blood group ABO involves three carbohydrate antigens: A, B, and H. A, B, and AB individuals express a glycosyltransferase activity that converts the H antigen to the A antigen (by addition of UDP-GalNAc) or to the B antigen (by addition of UDP-Gal), whereas O individuals lack such activity. It is expressed on endothelial cells, epithelial cells and granulocytes. Increased expression of this antigen has been observed on some tumor tissues such as gastric carcinomas, urothelial carcinomas, and colon carcinomas.

Description: The Lewis histo-blood group system comprises a set of fucosylated glycosphingolipids that are synthesized by exocrine epithelial cells and circulate in body fluids. The glycosphingolipids function in embryogenesis, tissue differentiation, tumor metastasis, inflammation, and bacterial adhesion. They are secondarily absorbed to red blood cells giving rise to their Lewis phenotype. This gene is a member of the fucosyltransferase family, which catalyzes the addition of fucose to precursor polysaccharides in the last step of Lewis antigen biosynthesis. It encodes an enzyme with alpha(1,3)-fucosyltransferase and alpha(1,4)-fucosyltransferase activities. Lewis blood group antigens are carbohydrate moieties structurally integrated in mucous secretions. Lewis antigen system alterations have been described in gastric carcinoma and associated lesions. Anomalous expression of Lewis B antigen has been found in some non-secretory gastric carcinomas and colorectal cancers.

CF® dyes are Biotium's next-generation fluorescent dyes. CF®405S is a blue fluorescent dye (Ex/Em 404/431 nm) with superior brightness compared to other blue dyes; it is also compatible with super-resolution imaging by SIM. Note: Conjugates of blue fluorescent dyes are not recommended for detecting low abundance targets, because blue dyes have lower fluorescence and can give higher non-specific background than other dye colors.

Description: Periphilin Antibody: Periphilin, known as PPHLN1 or gastric cancer antigen Ga50, plays a role in epithelial differentiation and contributes to epidermal integrity and barrier formation. It interacts with periplakin, a known precursor of the cornified cell envelope. Periphilin 1 is ubiquitously expressed and localizes to nucleus and cytoplasm. Existing as eight alternatively spliced isoforms, Periphilin is highly insoluble and contains a putative nuclear localization signal (NLS) within its N-terminal half, a prerequisite for the formation of insoluble complexes and a possible caspase recognition sequence and a potential nuclear export signal. Periphilin may play an important role in the nervous system.

Description: This antibody recognizes a protein of 36 kDa, identified as Thymidylate Synthase (TS) (EC 2.1.1.45). TS converts deoxyuridine monophosphate (dUMP) to deoxythymidine monophosphate (dTMP), which is essential for DNA biosynthesis. TS is also a critical target for the fluoropyrimidines, an important group of antineoplastic drugs that are widely used in the treatment of solid tumors. Both 5-FU and fluorodeoxyuridine are converted in tumor cells to FdUMP which inactivates TS by formation of a ternary covalent complex in the presence of the folate cofactor 5,10-methylenetetrahydrofolate. Expression of TS protein is associated with response to 5-fluorouracil (5-FU) in human colorectal, gastric, head and neck, and breast carcinomas.

Description: This MAb recognizes the peptide core of gastric mucin M1 (recently identified as Mucin 5AC). Its epitope is destroyed by beta-mercaptoethanol but not by periodate treatment. This mucin is present in primary ovarian mucinous cancer but usually absent in colorectal adenocarcinoma, thus showing an expression pattern opposite to MUC2. Together with a panel of antibodies, Anti-MUC5AC may be useful for differential identification of primary mucinous ovarian tumors from colon adenocarcinoma metastatic to the ovary. MUC5AC antibodies may also be useful for identification of intestinal metaplasia as well as in the identification of pancreatic carcinoma and pre-cancerous changes vs. normal pancreas.

Description: E-Cadherin is a classical member of the cadherin superfamily. The encoded protein is a calcium-dependent cell-cell adhesion glycoprotein composed of five extracellular cadherin repeats, a transmembrane region, and a highly conserved cytoplasmic tail. Mutations in this gene are correlated with gastric, breast, colorectal, thyroid, and ovarian cancers. Loss of function is thought to contribute to progression in cancer by increasing proliferation, invasion, and/or metastasis. The ectodomain of this protein mediates bacterial adhesion to mammalian cells, and the cytoplasmic domain is required for internalization. Identified transcript variants arise from mutation at consensus splice sites. Also, E-Cadherin has a potent invasive suppressor role and it is a ligand for integrin alpha-E/beta-7.

Description: Cathepsin E (CTSE) is a gastric aspartyl protease that functions as a disulfide-linked homodimer. It is a member of the Peptidase C1 family, and has a specificity similar to that of Pepsin A and Cathepsin D. CTSE is localized to the endoplasmic reticulum and Golgi apparatus, while the mature enzyme is localized to the endosome. It is expressed abundantly in the stomach, the Clara cells of the lung and activated B-lymphocytes, and at lower levels in lymph nodes, skin and spleen. CTSE is an intracellular proteinase that have a role in immune function, activation-induced lymphocyte depletion in the thymus, neuronal degeneration and glial cell activation in the brain. Futhermore, it probably involved in the processing of antigenic peptides during MHC class II-mediated antigen presentation.

Description: The Collagen triple helix repeat-containing protein (CTHRC1) is a glycosylated, secreted protein that is transiently expressed in the arterial wall in response to injury where it is thought to contribute to vascular remodeling by inhibiting collagen expression and deposition and promoting cell migration (1). CTHRC1 is highly expressed multiple human cancers, including pancreatic cancer where it plays a significant role in the progression and metastasis of the disease by regulating migration and adhesion of tumors cells (2). CTHRC1 overexpression can also be used as an independent prognostic marker in gastric cancer (3). Mutations at this locus have been associated with Barrett esophagus and esophageal adenocarcinoma (4).

Description: SRPX2 is a 465 amino acid secreted protein expressed in neurons of the brain, including the rolandic area. It has been suggested that SRPX2 enhances cell motility, migration and adhesion through FAK signaling in gastric and other cancer cells. Localized to the cytoplasm, SRPX2 is a ligand for uPAR (urokinase plasminogen activator), a receptor that is a crucial component of the extracellular plasminogen proteolysis system. SRPX2 may be responsible for rolandic seizures (RSs) associated with oral and speech dyspraxia and mental retardation (MR). The involvement of SRPX2 in these disorders suggests an important role for SRPX2 in the perisylvian region critical for language and cognitive development.

Description: The Trefoil Factor peptides (TFF1, TFF2 and TFF3) are expressed in the gastrointestinal tract, and appear to play an important role in intestinal mucosal defense and repair. TFF2 has been shown to inhibit gastrointestinal motility and gastric acid secretion. Recent data suggests a potential role for TFF2 in acute and chronic asthma (Nikolaidis, N.M. et al. Am. Journal Respir. Cell Mol. Biol. (2003) 4: 458-464). Recombinant Human TFF-2 is a 12.0 kDa polypeptide of 107 amino acid residues, which includes a 40-amino acid trefoil motif containing three conserved intramolecular disulfide bonds.

Description: SRPX2 Antibody: Sushi-repeat-containing protein X-linked 2 (SRPX2) is a neural gene functioning in the speech and language center of the human brain; mutations in this gene lead to epilepsy, speech dyspraxia, mental retardation and cognitive disorders. Recently, SRPX2 was found to be a novel mediator of angiogenesis and can act as a ligand for the urokinase-type plasminogen activator, a protein that can facilitate invasive migration of sprouting endothelial cells. SRPX2 is also overexpressed in gastric cancer, leading to increased phosphorylation levels of focal adhesion kinase and enhanced cellular migration and adhesion, suggesting that SRPX2 may be a potential target in the treatment of metastatic cancers.
