Description: G protein-coupled receptors (GPRs) are a protein family of transmembrane receptors that transmit an extracellular signal (ligand binding) into an intracellular signal (G protein activation). GPR signaling is an evolutionarily ancient mechanism used by all eukaryotes to sense environmental stimuli and mediate cell-cell communication. All of the receptors have seven membrane-spanning domains and the extracellular parts of the receptor can be glycosylated. These extracellular loops also contain two highly conserved cysteine residues which create disulfide bonds to stabilize the receptor structure. GPR105, also designated P2Y14, is widely expressed throughout many brain regions where it localizes to glial cells, and specifically co-localizes with astrocytes. GPR105 is upregulated when a tissue is immunologically challenged with lipopolysaccharide, leading to the theory that GPR105 may play an important role in modulating peripheral and neuroimmune function.

Description: Serine/threonine kinase which acts as an essential component of the MAP kinase signal transduction pathway. Plays an important role in the cascades of cellular responses evoked by changes in the environment. Mediates signaling for determination of cell fate such as differentiation and survival. Plays a crucial role in the apoptosis signal transduction pathway through mitochondria-dependent caspase activation. MAP3K5/ASK1 is required for the innate immune response, which is essential for host defense against a wide range of pathogens. Mediates signal transduction of various stressors like oxidative stress as well as by receptor-mediated inflammatory signals, such as the tumor necrosis factor (TNF) or lipopolysaccharide (LPS). Once activated, acts as an upstream activator of the MKK/JNK signal transduction cascade and the p38 MAPK signal transduction cascade through the phosphorylation and activation of several MAP kinase kinases like MAP2K4/SEK1, MAP2K3/MKK3, MAP2K6/MKK6 and MAP2K7/MKK7. These MAP2Ks in turn activate p38 MAPKs and c-jun N-terminal kinases (JNKs). Both p38 MAPK and JNKs control the transcription factors activator protein-1 (AP-1).

Description: Protein kinase C (PKC) is a family of serine- and threonine-specific protein kinases that can be activated by calcium and the second messenger diacylglycerol. PKC family members phosphorylate a wide variety of protein targets and are known to be involved in diverse cellular signaling pathways. PKC family members also serve as major receptors for phorbol esters, a class of tumor promoters. Each member of the PKC family has a specific expression profile and is believed to play a distinct role in cells. The protein encoded by this gene is one of the PKC family members. This kinase has been shown to be involved in many different cellular functions, such as neuron channel activation, apoptosis, cardioprotection from ischemia, heat shock response, as well as insulin exocytosis. Knockout studies in mice suggest that this kinase is important for lipopolysaccharide (LPS)-mediated signaling in activated macrophages and may also play a role in controlling anxiety-like behavior. Description: This mAb reacts with human Toll-like receptor 2 (TLR4). It is a member of the Toll-like receptor (TLR) family, which plays a fundamental role in pathogen recognition and activation of innate immunity. TLRs are highly conserved from Drosophila to humans and share structural and functional similarities. They recognize pathogen-associated molecular patterns that are expressed on infectious agents, and mediate the production of cytokines necessary for the development of effective immunity. The various TLRs exhibit different patterns of expression. This receptor has been implicated in signal transduction events induced by lipopolysaccharide (LPS) found in most gram-negative bacteria. Mutations in this gene have been associated with differences in LPS responsiveness. Multiple transcript variants encoding different isoforms have been found for this gene.

Description: Inhibitor of kappa light polypeptide gene enhancer in B-cells, kinase epsilon. The transcription factor NFκB is retained in the cytoplasm in an inactive form by the inhibitory protein IκB. Activation of NFκB requires that IκB be phosphorylated on specific serine residues, which results in targeted degra-dation of IκB. IκB kinase beta(IKKbeta), previously designated CHUK, interacts with IκB-betaand specifically phosphorylates IκB-betaon the sites that trigger its degradation, serines 32 and 36. The functional IKK complex contains three subunits, IKKbeta, IKKbetaand IKKgamma(also designated NEMO), and each appear to make essential contributions to IκB phosphorylation. IKK-i is a serine/threonine kinase that shares homology with IKKbetaand IKKbeta. IKK-i is pri-marily expressed in immune cells and is induced by lipopolysaccharide and by proinflammatory cytokines including TNFbeta, IL-1 and IL-6. Overexpression of IKK-i was shown to result in phosphorylation of IκBbetaon Ser32 and Ser36, and in NFκB activation, suggesting that IKK-i may act as an IκB kinase in the immune system.

Description: CD54, a member of the immunoglobulin superfamily of cell surface receptors and also known as Intercellular Adhesion Molecule-1 (ICAM-1), is a type I transmembrane glycoprotein that mediates important cell/cell interactions. CD54 is expressed on endothelial cells, dendritic cells, keratinocytes, and lymphocytes. Its expression is upregulated by inflammatory mediators such as IFN γ, IL-1, TNF-α, lipopolysaccharide, and phorbol esters. Endothelial CD54 contributes to the extravasation of leukocytes from blood vessels, particularly in areas of inflammation. CD54 on antigen-presenting cells contributes to antigen-specific T-cell activation, presumably by enhancing interactions between T cell and antigen-presenting cells. These adhesion reactions are mediated via the binding of CD54 to its major ligands, LFA-1 and Mac-1.

Description: Interleukin-22 (IL-22), also known as IL-10 related T cell derived inducible factor (ILTIF) was initially identified as a gene induced by IL-9 in mouse T cells and mast cells. IL-22 has been shown to activate STAT1 and STAT3 in several hepatoma cell lines and upregulate the production of acute phase proteins. IL-22 is produced by normal T cells upon anti-CD3 stimulation in humans. Mouse IL-22 expression is also induced in various organs upon lipopolysaccharide injection, suggesting that IL-22 may be involved in inflammatory responses. The functional IL-22 receptor complex consists of two receptor subunits, IL-22R (CRF29) and IL-10Rbeta (CRF24), belonging to the class II cytokine receptor family.

Description: Cooperates with LY96 and CD14 to mediate the innate immune response to bacterial lipopolysaccharide (LPS). Acts via MYD88, TIRAP and TRAF6, leading to NF-kappa-B activation, cytokine secretion and the inflammatory response. Also involved in LPS-independent inflammatory responses triggered by free fatty acids, such as palmitate. In complex with TLR6, promotes sterile inflammation in monocytes/macrophages in response to oxidized low-density lipoprotein (oxLDL) or amyloid-beta 42. In this context, the initial signal is provided by oxLDL- or amyloid-beta 42-binding to CD36. This event induces the formation of a heterodimer of TLR4 and TLR6, which is rapidly internalized and triggers inflammatory response, leading to the NF-kappa-B-dependent production of CXCL1, CXCL2 and CCL9 cytokines, via MYD88 signaling pathway, and CCL5 cytokine, via TICAM1 signaling pathway, as well as IL1B secretion.
