Description: Metabolism of inositol phospholipids by intracellular signaling mediators is fundamental to signal transduction in eukaryotic cells. PI-4,5-P2 (Phosphatidylinositol 4,5-bisphosphate; PIP2) can be synthesized by phosphorylation of PI-4-P by type I phosphatidylinositol phosphate kinase (PIP5K I), or phosphorylation of PI-5-P by type II PIPK (PIP4K II). PI-4,5-P2 regulation of cellular calcium levels involves its hydrolysis by Phospholipase C (PLC) to produce inositol 1,4,5-triphosphate (IP3) and diacylglycerol (DAG), which serve as second messengers in the import of calcium via IP3-sensitive ion channels and in the activation of PKC, respectively.

Description: MA3-500 detects PIP2 from human tissues. MA3-500 has been successfully used in ELISA, Immunoprecipitation, Immunofluorescence, and Western blotting procedures. By Western blot it detects a 67-kDa band representing MA3-500. The MA3-500 antigen is liposomes containing synthetic di-palmitoyl ptdlns(4,5)P2. Phosphatidylinositol 4,5-bisphosphate or PIP2 is an important second messenger for a number of signaling processes. When certain extracellular signals are received, Phosphilipase C cleaves phosphatidylinositol into DAG and PIP2. PIP2 has been shown to interact with actin structural proteins including profiling, gelsolin, and cofilin, and demonstrates regulatory function of actin polymerization. It has also been shown to regulate vesicular transport by interacting with proteins like ADP-ribosylation factor and phospholipase D. PIP2 has also been shown to significantly affect intracellular calcium concentrations, which also regulate a variety of activities.

Description: Synaptojanin 1 is a phosphoinositide phosphatase that regulates levels of membrane phosphatidylinositol-4,5-bisphosphate. As such, expression of this enzyme may affect synaptic transmission and membrane trafficking.Synaptojanin-1 is a phosphatidylinositol (4,5)-bisphosphate (PI (4,5)P2) that has a role in clathrin-coated pit dynamics Perera et al. (2006) [PubMed 17158794].

Description: This antibody is predicted to react with rat based on sequence homology. Phosphatidylinositol-4,5-bisphosphate, the precursor to second messengers of the phosphoinositide signal transduction pathways, is thought to be involved in the regulation of secretion, cell proliferation, differentiation, and motility. TPIP5K2A is one of a family of enzymes capable of catalyzing the phosphorylation of phosphatidylinositol-4-phosphate on the fifth hydroxyl of the myo-inositol ring to form phosphatidylinositol-4,5-bisphosphate. The amino acid sequence of this enzyme does not show homology to other kinases, but the recombinant protein does exhibit kinase activity. The protein is a member of the phosphatidylinositol-4-phosphate 5-kinase family.

Description: This Antibody targets PI4K2B in WB applications and shows reactivity with Human samples. The immunogen is recombinant fragment corresponding to a region within amino acids 198 and 481 of Human PI4K2B. Phosphatidylinositol 4-kinases (PI4Ks) phosphorylate phosphatidylinositol to generate phosphatidylinositol 4-phosphate (PIP), an immediate precursor of several important signaling and scaffolding molecules. PIP itself may also have direct functional and structural roles. PI4K2B is a primarily cytosolic PI4K that is recruited to membranes, where it stimulates phosphatidylinositol 4,5-bisphosphate synthesis (Wei et al. , 2002 [PubMed 12324459]).

Description: Phospholipase C beta 1 catalyzes the formation of inositol 1,4,5-trisphosphate and diacylglycerol from phosphatidylinositol 4,5-bisphosphate. This reaction uses calcium as a cofactor and plays an important role in the intracellular transduction of many extracellular signals. Its gene is activated by two G-protein alpha subunits, alpha-q and alpha-11. Two transcript variants encoding different isoforms have been found for its gene.

Supplier: Bioss
Description: This gene encodes a member of the phospholipase C family. Phospholipase C isozymes play critical roles in intracellular signal transduction by catalyzing the hydrolysis of phosphatidylinositol 4,5-bisphosphate (PIP2) into the second messengers diacylglycerol (DAG) and inositol triphosphate (IP3). The encoded protein functions as a tumor suppressor in several types of cancer, and mutations in this gene are a cause of hereditary leukonychia. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene. [provided by RefSeq, Dec 2011].

Catalog Number: (76077-800)
Supplier: Bioss
Description: PLCL1 is involved an inositol phospholipid based intracellular signaling cascade. Shows no PLC activity to phosphatidylinositol 4,5-bisphosphate and sphatidylinositol. Component in the dependent endocytosis process of GABA A receptor (By similarity). Regulates the turnover of receptors and thus contributes to the maintenance of GABA mediated synaptic inhibition. Its aberrant expression could contribute to the genesis and progression of lung carcinoma. Acts as an inhibitor of PPP1C.

Catalog Number: (103494-342)
Supplier: Affymetrix, Inc.
Description: Phosphoinositide-specific phospholipase C (PLC) plays a crucial role in the initiation of receptor mediated signal transduction through the generation of the two second messengers, inositol 1,4,5-triphosphate and diacylglycerol from phosphatidylinositol 4,5-bisphosphate. There are many mammalian PLC isozymes, including PLC beta1, PLC beta2, PLC beta3, PLC beta4, PLC gamma1, PLC gamma2, PLC delta1, PLC delta2 and PLC epsilon. PLC gamma1 is widely distributed in bronchiolar epithelium, type I and II pneumocytes and fibroblasts of the interstitial tissue. Actinregulatory protein Villin is tyrosine phosphorylated and associates with PLC gamma1 in the brush border of intestinal epithelial cells. Villin regulates PLC gamma1 activity by modifying its own ability to bind phosphatidylinositol 4,5-biphosphate. PLC gamma1 binds alpha1 beta1 Integrin and modulates alpha1 beta1 Integrin-specific adhesion. PLC gamma1 and Ca2+ play a direct role in VEGF-regulated endothelial growth, however this signaling pathway is not linked to FGF-mediated effects in primary endothelial cells.
