Description: May have a regulatory role in the membrane interactions during trafficking of synaptic vesicles at the active zone of the synapse. It binds acidic phospholipids with a specificity that requires the presence of both an acidic head group and a diacyl backbone. A Ca(2+)-dependent interaction between synaptotagmin and putative receptors for activated protein kinase C has also been reported. It can bind to at least three additional proteins in a Ca(2+)-independent manner; these are neurexins, syntaxin and AP2.

Description: Polycomb group (PcG) of proteins form the multiprotein complexes that are important for the transcription repression of various genes involved in development and cell proliferation. The protein encoded by RNF2 is one of the PcG proteins. It has been shown to interact with, and suppress the activity of, transcription factor CP2 (TFCP2/CP2). Studies of the mouse counterpart suggested the involvement of this gene in the specification of anterior-posterior axis, as well as in cell proliferation in early development. This protein was also found to interact with huntingtin interacting protein 2 (HIP2), an ubiquitin-conjugating enzyme, and possess ubiquitin ligase activity.

Description: The transcriptional repressing Polycomb-group (PcG) and transcriptional activating trithorax-group (trxG) genes of Drosophila are part of a cellular memory system responsible for the stable inheritance of gene activity. PcG proteins assemble into multimeric protein complexes, which are involved in maintaining the transcriptional repressive state of genes over successive cell generations. EED (embryonic ectoderm development) is the human homolog of Eed, a murine PcG gene homologous to the Drosophila homeotic gene, extra sex combs. The human EED protein is 99. 5% identical to the mouse EED protein and contains seven WD repeats, which are involved in protein-protein interactions. There are two human EED transcripts that contain a putative 407-nucleotide-long intron and give rise to two HEED protein isoforms, 535 and 494 amino acids in length. EED interacts in a highly specific manner, both in vitro and in vivo, with histone deacetylase (HDAC) proteins.

Description: DDA Antibody: DDA1 (DET1 and DDB1 associated 1), along with DET1 and DDB1 and a member of the UBE2E group of canonical ubiquitin-conjugating enzymes, comprise DDD-E2 complexes, which interact with multiple ubiquitin E3 ligases. One of these E3 ligases is Cul4-containing E3 ligase complex CRL4. Cells depleted of DDA1 spontaneously accumulate double-stranded DNA breaks in a similar fashion as Cul4A-, Cul4B-, or WDR23-depleted cells, suggesting that DDA1 interacts with the CRL4 complex and may be involved in the ubiquitination and subsequent proteasomal degradation of target proteins.

Description: Fanconi anemia (FA) is an autosomal recessive disorder characterized by bone marrow failure, birth defects and chromsomal instability (1,2). The FA Group C complementation group gene encodes the protein FANCC, which is located in both cytoplasmic and nuclear compartments. FANCC is expressed in a cell cycle-dependent manner, with the lowest levels at the G1/S boundary and the highest levels in the M-phase. The FANCC protein interacts with other FA complementation group proteins as well as non-FA proteins (3). A human a spectrin II (designated aSpIIs) acts as a scaffold to enhance interactions between FANCC and FANCA to form a nuclear complex (4,5). Another binding partner of FANCC is the BTB/POZ domain containing protein FAZF, which is a transcriptional repressor (6). In hematopoietic cells expressing mutant FANCC, PKR is constitutively phosphorylated and has increased binding affinity for double-stranded RNA (7,8), which suggests that FANCC indirectly suppresses the activity of PKR. These cells are also apoptotic and are hypersensitive to IFNg and TNFa (8). In addition, FANCC protein is involved in the activation of STAT1 through receptors for at least three hematopoietic growth and survival factors (8).
