Description: The Pax gene family of nuclear transcription factors comprises nine members that function during embryogenesis to regulate the temporal and position-dependent differentiation of cells. In addition, the family is involved in a variety of signal transduction pathways in the adult organism. Mutations in the Pax family of proteins have been linked to disease and cancer in humans. Pax-7 is a protein specifically expressed in cultured satellite cell-derived myoblasts. In situ hybridization reveals that Pax-7 is also expressed in satellite cells residing in adult muscle. A chromosomal aberration in the gene encoding Pax-7 causes rhabdomyosarcoma 2 (RMS2) (also called alveolar rhabdomyosarcoma).

Description: Antigen binding by the T-cell receptor (TCR) is one of the critical first steps in the immune response, triggering a cascade of signaling pathways that ultimately lead to T-cell activation. Screening a yeast two-hybrid screen of a human T-cell cDNA library with calcineurin, a protein phosphatase involved in multiple signaling pathways including T-cell activation, resulted in the identification of Carabin, a member of the TBC1 domain family of proteins, as a calcineurin-binding protein. Unlike other members of the TBC1 domain protein family which are thought to have a role in regulating cell growth and differentiation, further experiments demonstrated that Carabin is part of a negative regulatory loop for the intracellular TCR signaling pathway as well as an inhibitor of the Ras signaling pathway, suggesting that Carabin may also mediate crosstalk between calcineurin and Ras. Carabin antibody does not recognize TBC1D10A or TBC1D10B. Carabin is known to exist in multiple isoforms.

Description: Juvenile nephronophthsis, an autosomal recessive cystic kidney disease, is the primary cause for chronic renal failure in children. The gene for nephronophthsis type 1 (NPHP1) has recently been identified. Its gene product, nephrocystin, is a novel protein of unknown function, which contains a src-homology 3 domain.NPHP1 expression has been studied in mouse and human systems. Expression in the human adult is widespread but relatively weak. In contrast, expression in adult mouse is strong and localized to the testis. In situ hybridization to whole mouse embryo demonstrates widespread and consistently strong expression throughout all developmental stages.It is hypothesized that the NPHP1 gene product, nephrocystin, is involved in signaling processes at focal adhesions (cell and extracellular matrix contact) and/or adherens junctions (cell-cell contact). The hypothesis is based on the fact that most src-homology 3 -containing proteins are part of focal adhesion signaling complexes. Additionally, nephrocystin binds to p130(cas), a prominent participant in focal adhesion signaling.

Description: DISC1 Antibody: Disrupted in schizophrenia 1 (DISC1) is a candidate gene for susceptibility to schizophrenia. It was discovered through chromosomal analysis of a large Scottish family whose members exhibited schizophrenia and related psychiatric disorders. Through yeast two-hybrid screening, it was discovered that DISC1 interacts with many members of the centrosome and cytoskeletal system including MAP1A and Nudel. More recently, DISC1 has been found to regulate the transport of a complex containing Nudel, the lissencephaly-1 (LIS1) protein, and 14-3-3epsilon from neuronal cell bodies to the axons by the action of the microtubule-dependent directed motor protein kinesin-1, also known as KIF5A. Decreased expression of DISC1 in neurons caused an accelerated rate of neuronal integration, resulting in aberrant morphological development, suggesting that DISC1 plays a role in dendritic development and synapse formation. DISC1 has at least four known isoforms.

Description: FRMPD1 Antibody: The FERM and PDZ domain containing (FRMPD) protein family consists of four proteins that contain a FERM (Four-point-one, erzin, radixin, moesin) domain and at least one PDZ (PSD-95/Discs large/Zonula-occuldens-1) domain. FRMPD1 was initially identified through a yeast two-hybrid screening with Activator of G-protein signaling 3 (AGS3), a protein that acts to stabilize the GDP-bound conformation of Gai, as the bait. FRMPD1 binds to the tetratricopeptide repeat of AGS3 and stabilizes AGS3 in a membrane environment. Suppression of FRMPD1 in Cath.a-differentiated neuronal cells decreased the level of endogenous AGS3 in membrane fractions by ~50% and enhanced the inhibition of forskolin-induced increases in cAMP. Co-immunoprecipitation studies indicate that the interaction of AGS3 with FRMPD1 is mutually exclusive with the interaction of AGS3 with Gai3, suggesting that FRMPD1 may position AGS3 in the membrane where it can then interact with Galphai.

The rearrangement of chromosome 22 involves intron 8 of EWS and exon 1 of this gene creating a chimeric sequence containing the transactivation domain of EWS fused to zinc finger domain of this protein. Description: TWEAK is a TNF family member produced by IFN-gamma-stimulated monocytes and induces multiple cell death pathways including caspase-dependent apoptosis and cathepsin B-dependent necrosis. These pathways are mediated by the binding of TWEAK by fibroblast growth factor 14 (Fn14), a member of the TNF receptor superfamily. Yeast two-hybrid experiments have shown that FN14 will bind to the TNFR-associated factors TRAF1, TRAF2, TRAF3, and TRAF5, suggesting that the signaling pathway induced by TWEAK may be mediated by one or more of these adaptor molecules. Fn14 is induced during liver regeneration and is highly expressed in hepatocellular carcinomas. In addition, TWEAK induces liver progenitor cell proliferation, suggesting Fn14 may play a role in hepatocyte growth control and liver neoplasia.

Description: Autophagy, the process of bulk degradation of cellular proteins through an autophagosomic-lysosomal pathway is important for normal growth control and may be defective in tumor cells. It is involved in the preservation of cellular nutrients under starvation conditions as well as the normal turnover of cytosolic components. Beclin-1, a principal regulator of autophagosome formation, is in turn regulated by Ambra1. Ambra1 associates with Beclin-1 through a region near its center as determined by yeast two-hybrid assay. Null mutations in this gene in mice resulted in embryonic lethality with severe neural tube defects associated with autophagy impairment, accumulation of ubiquitinated proteins, unbalanced cell proliferation and excessive apoptotic death. Furthermore, down-regulation of Ambra1 in cultured cells though RNA interference decreased the level of rapamycin- and nutrient starvation-induced autophagy. Multiple isoforms of Ambra1 are known to exist.
