Description: Anti-PNL2 is a novel monoclonal antibody, which has recently been introduced as an immunohistochemical reagent to stain melanocytes and tumors derived therefrom. The antigen recognized by PNL2 is different from Melan A and gp100. Its epitope is not destroyed by digestion with neuraminidase i.e. its epitope id not glycosylated. Anti-PNL2 may be most useful because of its high sensitivity for metastatic melanoma (87%), as opposed to 76% for anti-HMB45 and 82% for anti-MART-1. Anti-PNL2 labels intra-epidermal nevi while the dermal component of compound nevi are largely non-reactive with anti-PNL2. Antibodies against PNL2, MART-1 (Melan A) and HMB45 stain most clear cell sarcoma cells and a few cells in angio-myolipomas and lymphangioleiomyomatosis. Anti-PNL2 is a useful antibody for the identification of melanomas and clear cell sarcomas. Differential diagnosis is aided by the results from a panel of antibodies, including antibodies against HMB45, MART-1, tyrosinase, and MiTF.

CLC3 expression is particularly abundant in neuronal tissue, while CLC4 expression is evident in skeletal and cardiac muscle as well as brain. Mutations in the gene encoding CLC5 lead to Dent’s disease, a renal disorder characterized by proteinuria and hypercalciuria. CLC6 and CLC7 are broadly expressed in several tissues including testis, kidney, brain and muscle.

Description: Dynamin is a 100 kDa GTP binding protein that was first identified as a protein able to bind microtubules. A mutation in the Drosophila gene shits, encoding a dynamin homologue, prevents synaptic vesicle recycling and causes temperature-sensitive paralysis, implicating dynamin in the formation of coated vesicles. Dynamin may mediate vesicle formation by assembling into a ring structure around the necks of invaginated coated pits that may eventually lead to the closure of the neck and formation of vesicles. Amphiphysin, dynamin, clathrin and AP2 form a complex during synaptic vesicle endocytosis, which is regulated by the phosphorylated state of different components in this complex. To date, three genes have been found to encode dynamin isoforms. Dynamin 1 is neuronal specific, Dynamin 2 is ubiquitously expressed and Dynamin 3 localizes to testis, brain and lung.
