Description: Isoform 1: Non-catalytic component of a structure-specific DNA repair endonuclease responsible for the 5'-incision during DNA repair. Responsible, in conjunction with SLX4, for the first step in the repair of interstrand cross-links (ICL). Participates in the processing of anaphase bridge-generating DNA structures, which consist in incompletely processed DNA lesions arising during S or G2 phase, and can result in cytokinesis failure. Also required for homology-directed repair (HDR) of DNA double-strand breaks, in conjunction with SLX4.

Description: Armadillo-like proteins are characterized by a series of armadillo repeats, first defined in the Drosophila 'armadillo' gene product, that are typically 42 to 45 amino acids in length. These proteins can be divided into subfamilies based on their number of repeats, their overall sequence similarity, and the dispersion of the repeats throughout their sequences. Members of the p120(ctn)/plakophilin subfamily of Armadillo-like proteins, including CTNND1, CTNND2, PKP1, PKP2, PKP4, and ARVCF. PKP4 may be a component of desmosomal plaque and other adhesion plaques and is thought to be involved in regulating junctional plaque organization and cadherin function. Multiple transcript variants encoding different isoforms have been found for this gene. (provided by RefSeq, Feb 2015).

Description: Apoptosis can be inhibited by a group of proteins called inhibitors of apoptosis (IAPs). These proteins contain a BIR (baculovirus IAP repeat) domain near the amino-terminus. The BIR domain can bind some caspases. Many members of the IAP family of proteins block proteolytic activation of caspase-3 and ~7. For example, XIAP, cIAP 1 and cIAP 2 appear to block cytochrome c-induced activation of caspase-9, thereby preventing initiation of the caspase cascade. Since cIAP 1 and cIAP 2 were first identified as components in the cytosolic death domain-induced complex associated with the TNF family of receptors, they may inhibit apoptosis by additional mechanisms.

Description: Tight junctions act as a semi-permeable barrier to the transport of ions, solutes, and water and are considered to function as a fence that divides apical and basolateral domains of plasma membranes. Tight junctions coordinate a variety of signaling and trafficking molecules regulating cell differentiation, proliferation, and polarity and contain a number of junctional proteins including Occludin, Claudins, junctional adhesion molecules (JAMs), as well as multiple scaffold proteins. Occludin, the first identified component of tight junction strands, is thought function as a signal transmitter in multiple signaling pathways and can associate with multiple kinases and phosphatases such as PI3-kinase and protein phosphatases 1 and 2A. At least two isoforms of OCLN are known to exist.

Description: Anti-PNL2 is a novel monoclonal antibody, which has recently been introduced as an immunohistochemical reagent to stain melanocytes and tumors derived therefrom. The antigen recognized by PNL2 is different from Melan A and gp100. Its epitope is not destroyed by digestion with neuraminidase i.e. its epitope id not glycosylated. Anti-PNL2 may be most useful because of its high sensitivity for metastatic melanoma (87%), as opposed to 76% for anti-HMB45 and 82% for anti-MART-1. Anti-PNL2 labels intra-epidermal nevi while the dermal component of compound nevi are largely non-reactive with anti-PNL2. Antibodies against PNL2, MART-1 (Melan A) and HMB45 stain most clear cell sarcoma cells and a few cells in angio-myolipomas and lymphangioleiomyomatosis. Anti-PNL2 is a useful antibody for the identification of melanomas and clear cell sarcomas. Differential diagnosis is aided by the results from a panel of antibodies, including antibodies against HMB45, MART-1, tyrosinase, and MiTF.

Description: The family of voltage-dependent chloride channels (CLCs) regulate cellular trafficking of chloride ions, a critical component of all living cells. CLCs regulate excitability in muscle and nerve cells, aid in organic solute transport and maintain cellular volume. The genes encoding human CLC-1 through CLC-7 map to chromosomes 7q32, 3q28, 4q32, Xp22.3, Xp11.23-p11.22, 1p36 and 16p13, respectively. CLC1 is highly expressed in skeletal muscle. Mutations in the gene encoding CLC1 lead to myotonia, an inheritable disorder characterized by muscle stiffness and renal salt wasting. CLC2 is highly expressed in the epithelia of several organs including lung, which suggests CLC2 may be a possible therapeutic target for cystic fibrosis. CLC3 expression is particularly abundant in neuronal tissue, while CLC4 expression is evident in skeletal and cardiac muscle as well as brain. Mutations in the gene encoding CLC5 lead to Dent’s disease, a renal disorder characterized by proteinuria and hypercalciuria. CLC6 and CLC7 are broadly expressed in several tissues including testis, kidney, brain and muscle.

Description: Ubiquitin is an abundant, highly conserved protein found in all eukaryotic cells either free or covalently attached to cellular proteins. The primary function of ubiquitin in mammalian systems is to clear abnormal, foreign, and improperly folded proteins by targeting them for proteosome degradation. UBE2D proteins, including UBE2D1 (ubiquitin-conjugating enzyme E2D1 or UBC5A), UBE2D2 (ubiquitin-conjugating enzyme E2D2 or UBC5B) and UBE2D3 (ubiquitin-conjugating enzyme E2D3 or UBC5C), are E2 ubiquitin-conjugating enzymes that catalyze the ubiquitination of I˚Bå in a phosphorylation and SCFB-TRCP-dependent manner. Specifically, E1 first transfers a ubiquitin residue to the E2 component (a UBE2D protein), and the UBE2D protein then associates with an E3 ubiquitin-protein ligase, which immediately transfers that residue to a protein that is targeted for degradation.

Description: The ubiquitin (Ub) pathway involves three sequential enzymatic steps that facilitate the conjugation of Ub and Ub-like molecules to specific protein substrates. The first step requires the ATP-dependent activation of the Ub C-terminus and the assembly of multi-Ub chains by the Ub-activating enzyme known as the E1 component. The Ub chain is then conjugated to the Ub-conjugating enzyme (E2) to generate an intermediate Ub-E2 complex. The Ub-ligase (E3) then catalyzes the transfer of Ub from E2 to the appropriate protein substrate. UBE2C, also designated UBCH10 in human, is an E2 ubiquitin conjugating enzyme for the anaphase promoting complex (APC), which coordinates mitosis and G1 by sequentially promoting the degradation of key cell-cycle regulators. UBE2C is overexpressed in many different types of cancers and may be a potential therapeutic target.

UBE2A and UBE2B are both nuclear and cell membrane proteins that have been found to interact with Rad18.

Description: Dynamin is a 100 kDa GTP binding protein that was first identified as a protein able to bind microtubules. A mutation in the Drosophila gene shits, encoding a dynamin homologue, prevents synaptic vesicle recycling and causes temperature-sensitive paralysis, implicating dynamin in the formation of coated vesicles. Dynamin may mediate vesicle formation by assembling into a ring structure around the necks of invaginated coated pits that may eventually lead to the closure of the neck and formation of vesicles. Amphiphysin, dynamin, clathrin and AP2 form a complex during synaptic vesicle endocytosis, which is regulated by the phosphorylated state of different components in this complex. To date, three genes have been found to encode dynamin isoforms. Dynamin 1 is neuronal specific, Dynamin 2 is ubiquitously expressed and Dynamin 3 localizes to testis, brain and lung.

Description: Payloads are the important components of antibody-drug conjugates (ADCs). The most commonly used ADC payloads are MMAE, DM1, and DXd. ADC pharmacokinetic (PK) studies include free payload analysis, conjugated antibody analysis, and total antibody analysis via ligand-binding assay (LBA) and LC-MS/MS assay. The anti-payload antibody is a useful reagent in PK assay for determining conjugated antibodies. It can be used as a capture antibody to determine the conjugated antibody in LBA assay or through immunocapture to enrich the ADC sample in LC-MS/MS assay. The DXd, also known as exatecan derivative, is a potent inhibitor of DNA topoisomerase I. Trastuzumab deruxtecan (T-DXd, DS-8201a or Enhertu®), composed of trastuzumab conjugated to DXd, is the first approved ADC for the treatment of breast cancer.
