Recombinant human SIK (1-303) was expressed by baculovirus in Sf9 insect cells using an N-terminal His tag. SIK is a protein kinase that is involved in regulating AMPK-related kinases (1). SIK may mediate the physiological effects of LKB1, including its tumour suppressor function. SIK is also involved in signaling by various proteins like STRAD, NUAK1, NUAK2, BRSK1, BRSK2, QIK, QSK, SIK, MARK1, MARK2, MARK3, MARK4 and MELK that are related to AMPK. Activation of SIK1 by phosphorylation on Thr322 can lead to an increase in the catalytic activity of sodium/potassium ATPase alpha subunit at the plasma membrane (2). This results in an increase in intracellular sodium in intact mammalian cells. ADP-Glo Kinase Assay is a luminescent kinase assay that measures ADP formed from a kinase reaction; ADP is converted into ATP, which is a substrate in a reaction catalyzed by Ultra-Glo Luciferase that produces light. The luminescent signal positively correlates with ADP amount and kinase activity. The assay is well suited for measuring the effects chemical compounds have on the activity of a broad range of purified kinases, making it ideal for both primary screening as well as kinase selectivity profiling. The ADP-Glo Kinase Assay can be used to monitor the activity of virtually any ADP-generating enzyme (e.g., kinase or ATPase) using up to 1mM ATP.
