Recombinant human MLK2 (1-446) was expressed by baculovirus in Sf9 insect cells using an N-terminal GST tag. MLK2 or mixed-lineage kinase 2 is a member of the serine/threonine kinase family, which activates MAPK8/JNK1 and MKK4/SEK1.

Recombinant human MLK2 (1-446) was expressed by baculovirus in Sf9 insect cells using an N-terminal GST tag. MLK2 or mixed-lineage kinase 2 is a member of the serine/threonine kinase family, which activates MAPK8/JNK1 and MKK4/SEK1. MLK2 itself can be phoshorylated and activated by JNK kinases. MLK2 is also known as mitogen-activated protein kinase kinase kinase 10 (MAP3K10). MLK2 functions preferentially on the JNK signaling pathway, which is involved in nerve growth factor (NGF)-induced neuronal apoptosis. MLK2 was isolated as a cDNA fragment from MKN28 gastric cancer cell library using degenerate PCR (1). MLK2 is highly expressed in brain, skeletal muscle and testis (2). ADP-Glo Kinase Assay is a luminescent kinase assay that measures ADP formed from a kinase reaction; ADP is converted into ATP, which is a substrate in a reaction catalyzed by Ultra-Glo Luciferase that produces light. The luminescent signal positively correlates with ADP amount and kinase activity. The assay is well suited for measuring the effects chemical compounds have on the activity of a broad range of purified kinases, making it ideal for both primary screening as well as kinase selectivity profiling. The ADP-Glo Kinase Assay can be used to monitor the activity of virtually any ADP-generating enzyme (e.g., kinase or ATPase) using up to 1mM ATP.
